細胞角蛋白17抗體；CK17抗體產(chǎn)品介紹：The protein encoded by this gene is a member of the keratin family. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. The type I cytokeratins consist of acidic proteins which are arranged in pairs of heterotypic keratin chains. Unlike its related family members, this smallest known acidic cytokeratin is not paired with a basic cytokeratin in epithelial cells. It is specifically expressed in the periderm, the transiently superficial layer that envelopes the developing epidermis. The type I cytokeratins are clustered in a region of chromosome 17q12-q21. This gene encodes the type I intermediate filament chain keratin 17, expressed in nail bed, hair follicle, sebaceous glands, and other epidermal appendages. Mutations in this gene lead to Jackson-Lawler type pachyonychia congenita and steatocystoma multiplex.

抗原修復(fù)的幾種常用方法（供參考）

1、 微波爐加熱：

在微波爐里加熱0.01*緩沖液（pH6.0）至沸騰后將組織芯片放入，斷電，間隔5-10分鐘，反復(fù)1-2次。根據(jù)玻片情況可參考采用微波加熱“3-5-3法”3分鐘溫火沸騰后靜止5分鐘，再溫火沸騰3分鐘即可。

適用的抗原：來源于人、大鼠、小鼠的組織標本；來源于其他動物種屬的組織標本。

2、 沸熱修復(fù)：

電爐或水浴鍋加熱0.01M*緩沖液（pH6.0）至95℃左右，放入組織芯片加熱10-15分鐘。

3、 高壓熱修復(fù)：

在沸水中加入EDTA（pH8.0）或0.01m*緩沖溶液（pH6.0）.蓋上不銹鋼鍋蓋，但不能鎖定。將玻片置于金屬染色架上，緩慢加壓，使玻片在緩沖液中浸泡五分鐘，然后將蓋子鎖定，小閥門將會升起來。10分鐘后除去熱源，置入涼水中，當小閥門沉下去后打開蓋子。此方法適用于較難檢測或核抗原的抗原修復(fù)。（骨及軟骨組織的標本選擇較溫和的微波加熱修復(fù)） 。

4、 酶消化方法：

常用0.1%*和0.4%*液。*使用前預(yù)熱37℃，消化時間約為5-30分鐘。

適用于被固定遮蔽的抗原：包括 Collagen.GFAP. Complement.Cytokeratin.c-erB-2.LCA.LN.等。

抗體修復(fù)方式:

修復(fù)液：0.01M 枸櫞酸（pH6.0）或者0.05M EDTA（pH8.0）

抗體修復(fù)方法：

方法1：沸水浴修復(fù)，將盛有修復(fù)液和玻片的燒杯置于沸水浴環(huán)境，保持外部沸騰狀態(tài)15min，自然冷卻至室溫；

方法2：微波修復(fù)，將盛有修復(fù)液和玻片的燒杯置于微波爐中，高火5min，?；?min，中火5min，自然冷卻至室溫。
