隨著時間的推移，色譜柱分離效果變差的原因有很多。其中，你使用的樣品對色譜柱后期的使用壽命有至關重要的作用。如果你使用的樣品比較“臟"。它將會導致固體顆粒雜質滯留在篩板上，色譜柱的篩板和柱頭發(fā)生堵塞。解決這類問題，我們強烈推薦你使用保護住或者過濾樣品。如果已經發(fā)生了柱頭或者篩板堵塞的情況，可嘗試沖洗色譜柱或者反相沖洗色譜柱。

It is important to remember that HPLC columns are consumables. Eventually the column you are using will have to be replaced.there are several parameters that you should keep note of in order to know when your column should be changed.

There are several reasons that a column will go bad over time. The samples you are using will play a big role . If using
" dirty " samples,a guard column is usually recommended. Samples can contain impurities that are absorbed onto the surface of the bonded phase or particles that could plug the frist of the column. In order to avoid these problems,flushing the column with a strong solvent(rinsing with water frist if using buffers)can help the column last longer or filter the samples. Storing the column in a strong solvent for long periods of time will also help. If problems still occur,putting the column on in the reverse direction could remove any potential blockage of the frits.
High temperatures, backpressures, and pH could also lead to column failure.Be sure to check the care and use sheets in your column box before running.
It is also important to note the column is not always the problem. Perisdic maintenance and cleaning of your instrument is very important in order to keep results consistent. Keeping a well-kept log book will save you time and money when trying to troubleshoot a problrm with your HPLC instrument.
